Capsules - Overview
Capsules are encapsulated medications coated with materials such as soluble gelatin or carrageenan. This prevents direct contact between the drug and the taste buds, which can cause nausea. Depending on the coating material, capsules are categorized as hard capsules and soft capsules. Hard capsules primarily contain powdered or granular medications, while soft capsules primarily contain liquid medications.
Types of Capsules
Hard Capsules
Hard capsules are normally employed for dry solid active ingredients. The capsules are manufactured in two halves – a body and cap – to assist with filling. Gelatin has been the material of choice for capsules because of the ability of a solution to gel and form a solid at a temperature just above room temperature.
The types of materials that can be loaded into empty capsules include:
Dry solids – Powders, granules, pellets or small tablets.
Semi-solids – Suspensions or pastes.
Liquids – Non aqueous liquids such as alcohols or oils.
Soft Gelatin Capsule
Soft gelatin capsules, also called soft gels, are thicker than hard gelatin capsules, and require special manufacturing equipment, either a rotary capsule machine or a dosator machine that uses the principle of drop formation to create the capsules. A rotary capsule machine can continuously manufacture the capsules and insert the fill. Typical production rates are between 25000 and 30000 capsules per hour.
Soft gel capsules are available in a variety of shapes and sizes e.g.
Cylindrical – 0.15 – 25 ml
Ovoid – 0.05 – 7 ml
Pear shaped – 0.3 – 5 ml
Spherical – 0.05 – 5 ml
Tubes – 0.3 – 5 ml

The taste-masking advantage alone can increase adherence, especially in pediatric or geriatric populations. To help reduce swallowing difficulties, the FDA recommends keeping capsule size at or below the standard 00 size.

Hard gelatin capsules consist of two cylindrical parts: a body and a cap, which fit tightly together. Empty hard gelatin capsules are filled with powders, microcapsules, granules, or pellets. They are used in over-the-counter medicinal products, such as vitamins, probiotics, and herbs. They are also used in prescription drugs that require mixing multiple components in a single capsule.
Hard and soft gelatin capsules primarily differ in their structure, filling method, and usage. For hard gelatin capsules, the manufacturing process is two-step – first the empty gelatin shell is created, then it is filled with the active substance. This process is technologically simple and cost-effective.
Soft gelatin capsules have a one-piece, flexible form and are produced in a single step using rotary die machines. The manufacturing process involves forming and filling the capsules in one continuous operation using rotating dies and molds, which results in a higher cost.Hard gelatin capsules are more versatile, making them a popular choice among pharmaceutical and supplement manufacturers. They are ideal when the active substance is in dry form or when the final product contains multiple ingredients.
Soft gelatin capsules offer more flexibility for liquid forms of medication and visual appeal. They are often transparent or semi-transparent, giving them a consumer-friendly appearance.
